A growing body of medical research suggests GLP-1 medications such as Ozempic, Wegovy and others may affect more than body weight, with studies pointing to possible benefits across the heart, kidneys, liver, lungs, joints, skin and even mental health.
Semaglutide, the active ingredient in Ozempic and Wegovy, has been studied for cardiovascular effects that researchers say go beyond what weight loss alone would explain. A major study published in The Lancet found that semaglutide reduced the risk of heart attacks, strokes and cardiovascular death among people with existing cardiovascular disease and overweight or obesity, with benefits observed across different body weights.
Joint pain has also drawn research interest. The STEP 9 clinical trial, published in the New England Journal of Medicine, found that semaglutide significantly reduced knee osteoarthritis pain and improved physical function in adults with obesity, though researchers are still working out how much of that effect comes from weight loss itself versus other mechanisms.
Kidney function is another area under study. GLP-1 drugs may help protect the kidneys through improvements in blood sugar, blood pressure and inflammation. In the FLOW trial, published in the New England Journal of Medicine, semaglutide reduced the risk of major kidney-related complications and cardiovascular death by 24% among patients with type 2 diabetes and chronic kidney disease.
Liver disease has emerged as another focus. MASH, or metabolic dysfunction-associated steatohepatitis, is a severe form of fatty liver disease marked by inflammation and liver cell damage. A clinical trial in the New England Journal of Medicine found semaglutide improved liver tissue findings in MASH patients, including reduced inflammation and scarring. In that trial, 62.9% of semaglutide recipients saw resolution of MASH without worsening fibrosis, compared with 34.3% of those on placebo.
Real-world data presented at the European Respiratory Society Congress has linked GLP-1 medications to fewer asthma attacks and COPD flare-ups, which researchers attribute to possible anti-inflammatory effects — though they caution more study is needed to confirm a direct respiratory benefit.
The brain also contains GLP-1 receptors in regions tied to appetite, emotion, motivation and reward response, which has led researchers to examine mental health and addiction. An observational study posted on the preprint server medRxiv linked semaglutide use to lower rates of substance-related and mood disorders. That study has not been peer-reviewed, and the authors do not claim the drug caused the reduction.
Skin conditions are also under investigation. A clinical trial published in JAMA Dermatology found that adding tirzepatide to an existing psoriasis treatment improved skin clearance in adults with moderate to severe psoriasis who were overweight or had obesity.
Researchers caution that it remains unclear how much of each benefit stems from weight loss itself, reduced inflammation, or other mechanisms specific to the drugs. Effects can vary by medication and by patient, and physicians say people considering these drugs should discuss the potential risks and benefits with a healthcare provider rather than rely on headline findings alone.
